Background: ADAMTS3

ADAMTS3 (A disintegrin and metalloprotease with thrombospondin motifs 3; also PC II-NP) is a 140-150 kDa member of the ADAMTS family of Zn metalloproteases. It is expressed by osteoblasts, chrondrocytes, myoepithelium and fibroblasts, and participates in collagen maturation. Together with ADAMTS2 and TS14, ADAMTS3 is known to process the N-terminus of procollagen. In particular, it cleaves the N-terminal globular region of collagen I and II, leading to fibril formation. Mature human proADAMTS3 is a secreted, 1185 amino acid (aa) glycoprotein. It is highly modular and contains a proregion (aa 38-201), a peptidase M12B domain (aa 256-460), a disintegrin region (aa 470-550), four TSP type I sequences (aa 551-1015), and a PLAC domain (aa 1015-1054). There are two potential splice variants. One shows a 38 aa substitution for aa 169-1205, while another shows an alternative start site at Met171. Over aa 19-712, human ADAMTS3 shares 91% aa identity with mouse ADAMTS3.
