In WB, this DNA Polymerase epsilon (3C5.1) antibody detects a single band of 220 kDa (in HeLa extracts) representing the catalytic subunit of DNA Polymerase epsilon. NB100-115 will immunoprecipitate, but only in the absence of salt and even then poorly. The observed molecular weight of the protein may vary from the listed predicted molecular weight due to post translational modifications, post translation cleavages, relative charges, and other experimental factors.

Background: DNA Polymerase epsilon catalytic subunit A

DNA Polymerase epsilon, also called DNA polymerase epsilon subunit 3 (POLE3) is a histone-fold protein which interacts with other histone-fold proteins to bind DNA in a sequence-independent manner. DNA Polymerase epsilon generates a complex with DNA polymerase epsilon subunit CHRAC1 and associates with naked DNA, which is then incorporated into chromatin, facilitated through the nucleosome-remodeling activity of ISWI/SNF2H and ACF1. DNA Polymerase epsilon is a component of the epsilon DNA polymerase complex consisting of four subunits: POLE, POLE2, POLE3 and POLE4, wherein, the interaction with POLE4 is a prerequisite for further binding with POLE or POLE2. DNA Polymerase epsilon interacts with CHRAC1, and together with CHRAC1, ACF1 and ISWI/SNF2H proteins, it forms the ISWI chromatin-remodeling complex, CHRA. DNA Polymerase epsilon histone-fold protein dimers combine within larger enzymatic complexes for facilitating the processes of DNA packaging, replication, and transcription.
