Immunocytochemistry/Immunofluorescence: B7-H2/ICOSLG Antibody (HK5.3) - Chimeric [NBP3-09018] - Immunofluorescence analysis of paraformaldehyde fixed mouse (Mus musculus) splenocytes on Shi-fix(TM) coverslips, permeabilized with 0.15% Triton stained with the chimeric rabbit version of HK5.3 (NBP3-09018) at 10 ug/ml for 1h followed by Alexa Fluor(R) 488 secondary antibody (1 ug/ml), showing weak membrane staining in a subset of cells. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP3-09018, DAPI, merged channels and an isotype control. The isotype control was stained with an anti-Fluorescein antibody followed by Alexa Fluor(R) 488 secondary antibody.ELISA: B7-H2/ICOSLG Antibody (HK5.3) - Chimeric - Azide and BSA Free [NBP3-09018]
ELISA: B7-H2/ICOSLG Antibody (HK5.3) - Chimeric [NBP3-09018] - ELISA Plate coated with B7-H2/ICOSLG-Fc fusion protein at a concentration of 5 ug/ml. A 3-fold serial dilution from 10,000 to 0.1 ng/ml was performed using NBP3-09018. For detection, a 1:4000 dilution of HRP-labelled goat anti-rabbit antibody was used.Applications for B7-H2/ICOSLG Antibody (HK5.3) - Chimeric - Azide and BSA Free
Application
Recommended Usage
Block/Neutralize
Optimal dilutions of this antibody should be experimentally determined.
ELISA
Optimal dilutions of this antibody should be experimentally determined.
Flow Cytometry
Optimal dilutions of this antibody should be experimentally determined.
Immunocytochemistry/ Immunofluorescence

Application Notes
This chimeric rabbit antibody was made using the variable domain sequences of the original Rat IgG2a format, for improved compatibility with existing reagents, assays and techniques.
This antibody has been used to detect murine ICOSL by flow cytometry. It is able to block ICOSL interacting with ICOSL and inhibit T-cell proliferation in a collagen type II (CII)-induced arthritis model.
